Optimising recombinant protein expression

Due to our modular cross-organism expression technology platform we can screen and optimise the expression of a given target protein in parallel, exploiting the individual strengths of the four most acknowledged host systems: E. coli, yeast, insect (Baculo-system) and mammalian cell. As a great benefit for our customers Trenzyme can deliver a fast feasibility answer, streamlining the project development and therefore ensuring a substantial time saving.

Custom recombinant protein expression and purification

Using state-of-the-art technologies and equipment, Trenzyme can realise recombinant protein expression and purification according to the needs of the customer. A vital network of collaboration partners enriches the sophisticated in-house possibilities, which also allows the development of complete upstream and downstream processes including exceptional analytical capabilities.

Stable mammalian cell line development for recombinant target expression and RNAi-mediated target knockdown

Trenzyme’s technology platforms for recombinant target expression or RNAi-mediated down-regulation in mammalian cells, the ExoIN and EndoOUT systems, are proven to be extremely valuable tools to rapidly generate highly individualised cell lines.

Due to the unique features of these systems, assay cell lines and protein production cells lines can be functionally validated very early on in the stable cell line development process.

  • Stringent coupling of a selectable marker with target expression (ExoIN) or with RNAi-mediated target knockdown (EndoOUT)
  • No need for clonal cell line due to extreme homogeneity within the whole population
  • Homogenous expression or knockdown after three days
  • Fast feasibility answer within one week
  • Stable and homogenous cell line available within two weeks
  • Tunable expression (ExoIN)
  • Maximum knockdown from single copy integration (EndoOUT)
  • ExoIN and EndoOUT are compatible with any cell line
  • ExoIN technology works with any target protein (secreted, membrane, nuclear and cytoplasmic)

Online order platform for customised cell lines

Based on the ExoIN technology, cytobox™ has established the first online order platform for customised CHO or HEK293-based cell lines. Simply paste the protein sequence of the target into the order form, select the parental cell line and enter the personal data to finish the order – ordering of a customised cell line is as easy as that.

cytobox will start with gene synthesis, including codon and DNA structure optimisation.

Fast online ordering of customised cell lines

Due to a highly streamlined process the final homogenous expression cell line will be ready within 30 business days. Even better, the timeline is guaranteed. The target expression is analysed by qPCR and cytobox guarantees success: if there is no expression there are no costs for the customer.

Every cell line will be delivered with a ready-to-use cytobox media pack. The validated media allows immediate culturing of the customised cell line without time loss due to media adaptation.

The very competitive price for a cytobox cell line includes everything from gene synthesis to the media starter pack.
